高性能菌种的设计与构建是绿色生物制造的关键🧑🏼✈️。以能量流和代谢流优化为核心的代谢工程理论在面对多层次动态调控生命系统时🧏🏻♀️👍🏻,只能采用设计-构建-测试-学习(DBTL)的自然进化法则,菌种构建时间长、人力物力消耗大。张翀课题组提出高通量基因型-表型关联数据(HT-GPA)驱动的设计理念,通过高通量基因型功能和工业表型特征数据测试与学习,低成本的获得高质量基因型-表型关联信息,指导高性能菌种的设计与构建。围绕上述理念🕵🏻♀️🛰,张翀课题组提出高通量并行DBTL策略,实现千量级基因型的低成本测试和数据学习;发明了基于微流控的菌种培养过程数据高通量测试技术与装备🙅🏻,显著提升菌种工业表型数据获取效率;在基因型功能和表型特征数据使能平台的基础上,提出了全基因组规模基因型-表型关联数据驱动的下一代细胞工厂系统创制新策略,成功构建一系列具有自主产权的高性能菌种并实现产业应用。
在Nature Chemical Biology,Science Advances,Nature Communications🍦,Nucleic Acid Research, Metabolic Engineering和Biotechnology Bioengineering等期刊发表论文60余篇;申请中国发明专利19项🆙,授权12项,其中8项实现成果转化;牵头制定5项国家标准;微生物育种装备成功实现商业化销售并出口海外🪠,系列自主知识产权菌株实现在传统发酵和生物医药行业应用,并创造显著经济和社会效益。
